The five mechanisms studied in longevity research

Cellular energy declines with age as NAD+ availability falls, which is why NAD+ is central to the category and studied at 25–100 mg, 3–5× weekly. Mitochondrial signalling is addressed by MOTS-C, investigated for AMPK activation and mitochondrial biogenesis — the pathway most consistently linked to longevity endpoints in the literature. Why longevity protocols run long and low

Unlike acute-injury models, longevity research favours extended, lower-dose protocols: typically 6–12 weeks with maintenance phases continuing afterwards, on the reasoning that the pathways involved are chronic rather than episodic. GHK-Cu is a notable exception in that it requires no cycling at all. Because the compounds here address non-overlapping mechanisms, they are the most frequently combined group in the range.

Which anti-aging peptide should be studied first?

NAD+ and MOTS-C form the standard entry pairing because they address cellular energy and mitochondrial signalling — the foundational pathways. GHK-Cu is added when structural or skin ageing is also an endpoint.

What is the difference between NAD+ and MOTS-C?

NAD+ is a coenzyme required for energy metabolism and DNA repair, and its availability declines with age. MOTS-C is a mitochondrial-derived peptide that signals through AMPK to influence how cells regulate energy. One supplies a substrate; the other modulates the pathway.

Do these compounds stop ageing?
