Mechanism

How it works

Bacteriostatic water is not a peptide. It is a diluent — water for injection with a single additive, benzyl alcohol, at 0.9% by volume, which is 9 mg per millilitre.

The additive is the entire point. Benzyl alcohol is bacteriostatic rather than bactericidal: it inhibits the multiplication of organisms introduced into the vial rather than sterilising a contaminated solution. It does not rescue a compromised vial; it suppresses growth of the small numbers of organisms a stopper penetration can carry in, so that a vial entered several times over a working period does not become a culture. Its activity is attributed to disruption of microbial membrane integrity.

Sterile water for injection contains no preservative. It is sterile at the point of manufacture and remains so only until the stopper is first penetrated, after which it is single-use. For a lyophilised peptide reconstituted once and drawn down repeatedly over days, that is the wrong diluent — which is why bacteriostatic water is the convention for repeat-entry research vials, and sterile water the convention for single-use preparations and for assays where benzyl alcohol would interfere.

Two further distinctions matter. Bacteriostatic water is not saline: it contains no sodium chloride and is not isotonic. And it is not universally compatible. The Bacteriostatic Water for Injection USP labelling is explicit that benzyl alcohol-containing solutions must not be used in neonates, and that only preservative-free sterile water should be used for that population. The laboratory parallel is assay interference: benzyl alcohol is an organic solvent at low concentration and can affect chromatographic, cell-based and protein-binding assays.

Preparation & dosage

The following is general laboratory technique for reconstituting lyophilised research peptides. It is not a protocol for preparing material for administration to humans or animals.

Begin with both vials at ambient temperature. A cold peptide vial condenses atmospheric moisture onto the cake, and moisture is the principal cause of incomplete dissolution.

Decide the volume before piercing anything. Concentration is mass divided by volume: a 10mg peptide vial reconstituted with 2 mL gives 5 mg/mL; the same vial with 1 mL gives 10 mg/mL. Choose the volume that puts the working concentration where the model needs it.

Swab both stoppers with 70% isopropyl alcohol and allow them to dry. Alcohol that is still wet is not doing anything.

Draw the intended volume of bacteriostatic water. Insert the needle into the peptide vial at an angle so the stream runs down the inner glass wall. Do not aim it at the lyophilised cake — the mechanical force of a direct stream is a common cause of degradation. Release the diluent slowly.

Withdraw the needle and let the vial stand. Most cakes dissolve within a few minutes. If material remains, roll the vial gently between the palms or swirl it. Do not shake, vortex or invert vigorously; foaming indicates denaturation at the air-liquid interface and is the visible sign of a peptide being damaged.

Inspect the solution. It should be clear and free of particulates. Copper-containing peptides such as GHK-Cu are the intended exception, giving a blue to blue-violet solution.

Label the vial with the compound, concentration and reconstitution date, and store refrigerated at 2–8 °C, protected from light. Aliquot before freezing rather than freeze-thawing repeatedly.

What is the difference between bacteriostatic water and sterile water for injection?

One additive. Both are sterile, non-pyrogenic water; bacteriostatic water also contains 0.9% benzyl alcohol as a preservative. That preservative inhibits microbial growth across repeated stopper penetrations, so a vial can be entered more than once. Sterile water for injection has no preservative and is a single-use material once penetrated.

How much bacteriostatic water should be added to a peptide vial?

There is no universal figure — the volume sets the concentration, and the concentration should suit the research model. Concentration equals the mass in the vial divided by the volume added: a 10mg vial with 2 mL gives 5 mg/mL. Where a supplier specifies a volume, that specification takes precedence.

Why should the vial not be shaken?

Shaking drives peptide molecules to the air-liquid interface, where they unfold and aggregate. Foaming is the visible sign. The result is loss of assayed potency with no change in appearance once the foam settles, which is why the damage goes unnoticed. Swirl or roll instead, and allow time for dissolution.

Which size should be selected, and can one vial serve several peptides?

Match the vial to the work in hand. A 3ml vial suits a single reconstitution; 5ml and 10ml suit several within the preservative's in-use period. One vial can be entered repeatedly — that is the purpose of the preservative — provided the stopper is swabbed with 70% isopropyl alcohol before every entry and a fresh needle used each time. Never return drawn diluent to the vial.
